The plot revolves around a neighborhood being terrorized by a sentient haunted house during Halloween. The film features the voices of Mitchel Musso, Sam Lerner, Spencer Locke, Steve Buscemi, Maggie Gyllenhaal, Kevin James, Nick Cannon, Jason Lee, Fred Willard, Jon Heder, Catherine O'Hara, and Kathleen Turner. With the house destroyed, Constance’s spirit is released and able to go on to Heaven.

Judging from her photos that Jenny and DJ find in her and Nebbercracker's house, it was shown that her extreme obesity was due to her love of food. Whether this was true or not (or merely speculated), her size disgusted many people, and even children. But Constance was just a normal human who, after her death, became a vengeful spirit that possessed her own house. In fact, she seems to have an adept mastery at controlling non-human matter around the house. When Horace rescues Constance from the sideshow (again, Monster House wants you to believe that fat people need thin people to swoop in and save the day), he hitches the cage she’s trapped in to his truck and tows her away.

But little did he know, even though Constance's body died, her spirit came to inhabit the very house she wanted to live in due to her unwillingness to leave her husband. The abuse she suffered in life led to her distrusting anyone that would come near her, especially children, and as a result, she became a bloodthirsty and murderous monster house, attacking anyone that dared to approach her, except for Horace himself. Things were especially hectic for him on Halloween, where children would commit pranks, as he would take precautions by yelling at the kids to stay away from his house for their safety. On one Halloween day, after the house had been half-way built, two trouble-making children came by to throw rocks at it. As Horace has just started to take apart the cage his wife was formerly forced to live in with an ax, Constance came to him and begged him to help stop the kids from further damaging their property.

Whilst managing to calm Constance down, Mr. Nebbercracker laments over the fact that he has failed to keep his wife's wrath from hurting others and turning her into what she is now; a true monster. This saddens the house and provides Mr. Nebbercracker an opportunity to end his wife's suffering using a stick of dynamite. The house does not take this well and begins to roar ferociously, only for Mr. Nebbercracker to calm her down by telling her he has always done what is best for her. After scraping a match against the wood to light it, the house gasps in shock and Mr. Nebbercracker prepares to set the dynamite on fire with the match, allowing it to be the right thing to do.
Here, Jenny stated that the main hall was deformed into some kind of mouth cavity where the opened hole acts like a throat, the rug acts as a tongue, deformed woods around the door acts as teeth, while the hanged lamp acts as a uvula. Later on, when DJ and Chowder sought to investigate the house, Chowder, who does not believe in the strange things that occurred inside the house, starts to fool around on the house's lawn. They discovered Bones's empty beer bottle, but dismissed it and tossed it aside, unaware that the possessed grass on the lawn pulled it underground. Everything seems normal, until Chowder pressed the bell, where the house awakened. The door suddenly opens, revealing that its interior fractures and shifts into a mouth-like cavern.

When they entered it, they discovered Constance's corpse that was covered by cement layer. Around her dead body, they found some decayed roses, heart-shaped boxes of chocolates and photos that people would normally place on the grave of their loved ones. Before they could drive away, however, the house groans on them, luring both the police officers closer.
Mixing humor, horror, and a poignant message of the dangers of bullying and needs for acceptance, the film also blended Gothic elements with a kids’ friendship caper. Nebbercracker is heartbroken over the loss of his house and his wife, but is grateful to DJ, Chowder, and Jenny for freeing them from all these years of imprisonment. Nebbercracker stands up to Constance and realizes how much trouble she has caused recently, making her feel remorse for her actions. Nebbercracker then resolves to put Constance out of her misery by blowing her up with a dynamite. Infuriated by this “betrayal”, Constance grabs her husband, intent on killing him too. Before she has the chance, Chowder fights her off using the excavator from the adjoining construction site.
Oblivious with what has lured them, the two officers decided to investigate the house against the kids' wishes. Officer Lister taunts the tree, which suddenly, prompts it to move and grab his gun then grab his leg. Things suddenly became worse as the house reveals its true nature and devours Officer Landers alive while sadistically playing with Officer Lister that it grabbed via branches, before it eventually tossed him into its mouth.

Rather than freeing her and letting her ride in the truck with him like a human being, he treats her like so much cargo or like a wild animal that has to be caged up to protect the rest of the world from it. Horace attempts to calm her down by saying that they are just children merely joking because it was Halloween and vowing to never let anyone hurt her again as long as he's here. But just after that, one of the boys throws an egg at Constance, making her lose her temper. She grabs the ax from Horace's hands and attempts to attack the kids with it as they continue to throw eggs at her, while Horace attempts to stop her. During the scuffle, Horace falls to the ground but is okay while Constance loses her footing and stumbles towards a hole into the house's basement. She tries to save herself by grabbing the lever of the cement mixer, but her hand slips and falls into the pit of what would become the basement.
The children, left to their own devices, must conquer the evil that, all but unnoticed, flourishes in their midst. The house remains still until morning, though DJ and Chowder continue to watch it from DJ's room for hours even with Zee's interference and resort to pee in bottles so they will not miss any of the possessed house's activities. At that day, the children threw rocks at her, in which she responded by asking Mr. Nebbercracker for help.
As they were waiting, the trio imagines what would happened if many children come to that house due to tonight is Halloween. Just then, the house moves again and taunts Chowder by bouncing his basketball in the doorway that was then marked with a Jack-O-Lantern's face, which Chowder perceives that the house would start the real killing spree tonight. The battle between Chowder and Constance, causing them both to fall down into the bed of the empty lake; the house falls apart in the process. But, as DJ, Jenny, and Chowder rejoice, Constance reassembles the house, making it look like a massive misshapen bundle of wood, brick, and shingles.
There, they discovered Mr. Nebbercracker's old explosives and photographs depicting Mr. Nebbercracker when he was young alongside his wife Constance when she was alive. DJ stated that Zee and Bones had a conversation about Mr. Nebbercracker's past life and assumed that he devoured Constance, much to Jenny's disgust. However, this nearly made the house awaken as the house' main hall turned into a mouth-like cavern before spitting a large amount of water. It then had a beam of light to search the house' main hall where the beam came from one of the windows.
She lost her balance while trying to chase them away, falling to her death into the basement of their still unfinished house along with some fresh cement. Having become part of the foundation, she possessed and physically became the house, desiring to kill all the children of the neighborhood. Chowder is constantly talking about food—he looks forward to getting candy when they go trick-or-treating, and he mentions his favorite taco stand when he tries to hit on Jenny, the skinny girl who of course becomes DJ’s love interest rather than Chowder’s. Skinny characters call Chowder things like “Porky” or “pork chop.” When he’s not talking about food or being insulted for his weight, he’s making mistakes that show how stupid and clumsy this movie thinks fat people are. When the kids attempt to fool the house using a decoy trick-or-treater (a vacuum cleaner disguised as a child), Chowder has to be reminded to plug the vacuum in to make it move forward. So when I saw that venomous bullying in a movie meant to introduce kids to the horror genre—a genre for the outcasts, for those who don’t fit in, for those who are desperately seeking a welcoming home—I felt like I’d been slapped in the face.
